Basic fuchsin should be completely dissolved before adding sodium metabisulfite and HCl. There are a number of different recipes for Schiff's type solutions but most specify 1 gm of basic fuchsin in hot water (e.g. 80-100° C). The container should be closed tightly after adding the sodium tetrasulfate and the HCl and then permitted to stand a few hours in the dark. When ready, the solution should have a strong sulfur odor.
There is some variation in color of the final solutions and this has been the cause of considerable discussion as to what color is "right" and which colors are not right. Pale colored solutions are usable so long as they are not pink colored and control slides yield results as one would expect from them.
A slight orange color is generally perceived to be caused by impurities in the dye (acridine orange).
