Used widely worldwide as an electron dense metal stain for ultra-thin sections, lead solutions are frequently employed after uranyl staining to produce high contrast and to also stain many cellular and tissue components. Indeed the Reynolds stain is widely used as reported in Reynolds, J. Cell Bio 17, 208 (1963). However, carbon dioxide can be a problem with its use. This is also one of the key components of "Sato lead", sometimes called "Sato's lead citrate". Actually there is quite a bit of additional information available about different protocols and procedures. One should be quite exact in the mixing of the lead citrate for making the final staining solution.
