Some prototype staining protocols

There are probably about as many published protocols for the staining as there are researchers doing immunogold work on L R White embedded samples. But a "generalized" protocol could include
a) staining of the
immunogold
labeled grids with 3-5%
uranyl acetate
for five to ten minutes followed by
b) thorough washing with distilled water after the uranyl acetate treatment
c) take a few pellets of
sodium hydroxide
in a covered petri dish containing
lead citrate
and incubate the grids for ten minutes
d) air dry for 5-10 minutes before insertion into the vacuum of the TEM for viewing.
We publish this "generalized" protocol because there are numerous variations in existence. In general, these protocols are traceable back to the publications of Venable and Coggeshall.
This type of protocol gives very good contrast, and one should have no difficulty seeing the immunogold labeling in the section.
