Selection of particle size
Which particle size should be selected?
For the selection of the particle size, it is recommended that this be determined
by the magnification to be used in the electron microscope. For the highest
labeling intensity, however, the smaller gold particles are preferred. For
gold probes 5 nm or less, a combination of gold labeling with silver
enhancing will yield larger size particles with high labeling intensity.
For those researchers just starting out, if you are going to try reproducing
someone else's work to validate your technique, then obviously, you will want
to use the same size as was used in the publications. But if that is not the
case, and you are starting out from scratch, we recommend starting with 10 nm
gold particles size for observation at a whole range of magnifications.
